Alpha-Ketoglutarate
Promising mouse lifespan data, honest gaps in the human evidence
Alpha-ketoglutarate (AKG) is a molecule your body already makes — it's a central intermediate in the Krebs cycle, the metabolic pathway every cell uses to convert food into usable energy. It's also a required cofactor for a family of enzymes (Fe(II)/α-ketoglutarate-dependent dioxygenases) involved in collagen synthesis and in regulating DNA methylation, the chemical tagging system that changes as cells age.
What turned AKG into a longevity-supplement ingredient wasn't a human trial — it was a 2020 mouse study showing that late-life supplementation with calcium alpha-ketoglutarate (Ca-AKG) extended median lifespan and meaningfully reduced frailty. That's a genuinely striking result. It is not, on its own, proof that the same thing happens in people, and a dedicated human trial to test exactly that question is still in progress as of this writing.
Lifespan & Frailty (Mouse Data)
In aged mice, late-onset Ca-AKG supplementation significantly increased median lifespan and survival in females, and cut frailty scores by roughly 40-46% in both sexes — driven partly by reduced inflammatory cytokines.
DNA Methylation & Epigenetic Age
AKG is a required cofactor for TET enzymes and other dioxygenases that regulate DNA methylation patterns — the mechanism the ongoing human trial is testing directly, by measuring whether AKG lowers "biological age" on methylation clocks.
Collagen & Connective Tissue
AKG is a cofactor for prolyl-4-hydroxylase, the enzyme that stabilizes collagen's structure during synthesis — part of why it's also being studied in bone and cartilage research contexts.
Anti-Inflammatory Signaling
The mouse lifespan study's proposed mechanism centers on AKG inducing IL-10, an anti-inflammatory cytokine, which may help explain the reduced frailty seen alongside the lifespan extension.
- Take with food; calcium-bound forms are absorbed better with a meal
- Don't expect the mouse lifespan effect to translate directly — treat this as an emerging, not established, longevity ingredient
- Check total calcium intake if also taking a separate calcium supplement
- Often stacked with NAD+ precursors (NMN, NR) in longevity protocols, though the two haven't been studied together in a human trial
A 2020 study in Cell Metabolism found that late-onset dietary Ca-AKG supplementation in aged mice significantly increased median lifespan in females (by 16.6-19.7% across two cohorts) and reduced frailty scores by 40-46% in both sexes, with reduced inflammatory cytokines as the proposed mechanism. Human confirmation is in progress: the ABLE trial, a randomized, double-blind, placebo-controlled study in 120 middle-aged adults, is currently testing whether 1g/day of sustained-release Ca-AKG lowers DNA methylation age over 6 months — as of now, only the study protocol has been published, not results.
