Overview

Alpha-ketoglutarate (AKG) is a molecule your body already makes — it's a central intermediate in the Krebs cycle, the metabolic pathway every cell uses to convert food into usable energy. It's also a required cofactor for a family of enzymes (Fe(II)/α-ketoglutarate-dependent dioxygenases) involved in collagen synthesis and in regulating DNA methylation, the chemical tagging system that changes as cells age.

What turned AKG into a longevity-supplement ingredient wasn't a human trial — it was a 2020 mouse study showing that late-life supplementation with calcium alpha-ketoglutarate (Ca-AKG) extended median lifespan and meaningfully reduced frailty. That's a genuinely striking result. It is not, on its own, proof that the same thing happens in people, and a dedicated human trial to test exactly that question is still in progress as of this writing.

AKG is a cofactor for prolyl-4-hydroxylase, the enzyme that stabilizes collagen's structure during synthesis — part of why it's also being studied in bone and cartilage research contexts.

Anti-Inflammatory Signaling

The mouse lifespan study's proposed mechanism centers on AKG inducing IL-10, an anti-inflammatory cytokine, which may help explain the reduced frailty seen alongside the lifespan extension.

Does alpha-ketoglutarate actually extend lifespan in humans?
We don't know yet. The lifespan-extension evidence comes from a 2020 mouse study. A human trial (the ABLE study) is currently underway testing whether Ca-AKG lowers biological age on DNA methylation clocks, but as of now it has published only its study design, not results. Mouse data is a real signal, not proof of a human effect.
What form of AKG should I take?
Calcium alpha-ketoglutarate (Ca-AKG) is the form used in both the mouse lifespan study and the ongoing human trial, since free AKG has poor oral bioavailability. Most consumer supplements use this same calcium-bound form.
Is this the same AKG bodybuilders used in the 1990s?
Same base compound, but the older bodybuilding use (often ornithine alpha-ketoglutarate, OKG) was marketed for muscle recovery and growth hormone claims with thin evidence. The current longevity research uses standalone calcium AKG and tests an entirely different outcome — biological aging markers, not muscle building.
