Nicotinamide Riboside
A well-studied NAD+ precursor with real human trials on safety and blood NAD+ levels
Nicotinamide Riboside (NR) is a form of vitamin B3 and one of two popular precursor molecules — the other being NMN — that your body converts into NAD+, a coenzyme every cell needs to produce energy and repair DNA. NAD+ levels decline measurably with age, and the premise behind NR supplementation is straightforward: replenish the raw material to help restore more youthful NAD+ levels.
NR has one of the more mature human research bases in the "longevity supplement" category, including a well-designed long-term safety and dosing trial. What it hasn't been shown to do — and what no NAD+ precursor has been shown to do — is extend human lifespan; that evidence exists only in mice so far. It's best understood as a well-tolerated, biomarker-backed metabolic support supplement, not a proven anti-aging treatment.
Raises Blood NAD+ Levels
A randomized, placebo-controlled crossover trial found NR reliably and significantly raised blood NAD+ in healthy middle-aged and older adults.
Well-Established Safety Profile
An 8-week dose-ranging trial found doses up to 1,000mg/day well tolerated, with no significant increase in adverse events versus placebo.
No Adverse Cholesterol Effect
The same dose-ranging trial found NR did not elevate LDL cholesterol or disrupt related metabolic markers, even at the highest dose tested.
GRAS-Recognized Ingredient
Recognized as Generally Recognized as Safe (GRAS) by the FDA for use in food products, reflecting a favorable regulatory safety review.
- Treat it as metabolic-support, not a proven longevity guarantee — the human evidence supports raised NAD+, not extended lifespan
- Take with or without food — trials haven't shown a meaningful absorption difference either way
- Don't pay a premium for "proprietary" NR forms without independently verified human data behind the specific claim
- Talk to your doctor before starting, especially if you're on other medications, since long-term data beyond several months is still limited
An 8-week randomized, double-blind, placebo-controlled trial in healthy overweight adults found that 100mg, 300mg, and 1,000mg daily doses of NR dose-dependently raised whole blood NAD+ by 22%, 51%, and 142% respectively, with no significant difference in adverse events between NR and placebo groups and no negative effect on LDL cholesterol. A separate crossover trial in healthy middle-aged and older adults confirmed NR was well tolerated and effectively raised blood NAD+ levels.
How Nicotinamide Riboside Compares
| Ingredient | Best For | Key Difference |
|---|---|---|
| Nicotinamide Riboside | NAD+ Support, Longest Human Track Record | Most human trial data of the NAD+ precursors |
| NMN | NAD+ Support, Metabolic Function | One fewer conversion step to NAD+; a trial found improved muscle insulin sensitivity |
| Resveratrol | Sirtuin Activation Support | Often paired with NAD+ precursors, though human co-supplementation data is limited |
| CoQ10 | Cellular Energy, Different Pathway | Supports mitochondrial energy production via a separate mechanism from NAD+ |
