A nucleotide building block that supplies raw material for new brain synapses
Uridine is a nucleotide -- one of the basic building blocks of RNA -- but its supplement interest has nothing to do with genetics. In the brain, uridine is a rate-limiting precursor for synthesizing phosphatides, the specialized fats that make up the membranes of synapses, the junctions where neurons communicate. MIT researcher Richard Wurtman and colleagues characterized uridine as one of three circulating nutrients -- alongside choline and the omega-3 fat DHA -- that together supply the raw materials brain cells need to build new synaptic membrane and, in animal studies, new dendritic spines.
That's a genuinely interesting, mechanistically specific story, and it's part of why uridine shows up in nootropic stacks alongside citicoline and omega-3s. The honest caveat is that most of the strongest data -- on actually growing new synapses and improving memory -- comes from animal research. Human evidence exists, but it's smaller in scale and earlier-stage than the animal work, which is worth knowing before treating uridine as a settled cognitive enhancer.

A controlled human study using 31-phosphorus magnetic resonance spectroscopy found just seven days of uridine supplementation measurably increased brain membrane phospholipid precursors in healthy adults -- direct evidence the proposed mechanism is active in people, not just animals.

A small open-label case series in adolescents with bipolar depression found uridine associated with reduced depressive symptoms and a notably rapid onset -- promising, but explicitly described by the researchers as needing placebo-controlled confirmation before any clinical conclusions.
