A well-studied NAD+ precursor with real human trials on safety and blood NAD+ levels
Nicotinamide Riboside (NR) is a form of vitamin B3 and one of two popular precursor molecules — the other being NMN — that your body converts into NAD+, a coenzyme every cell needs to produce energy and repair DNA. NAD+ levels decline measurably with age, and the premise behind NR supplementation is straightforward: replenish the raw material to help restore more youthful NAD+ levels.
NR has one of the more mature human research bases in the "longevity supplement" category, including a well-designed long-term safety and dosing trial. What it hasn't been shown to do — and what no NAD+ precursor has been shown to do — is extend human lifespan; that evidence exists only in mice so far. It's best understood as a well-tolerated, biomarker-backed metabolic support supplement, not a proven anti-aging treatment.
A randomized, placebo-controlled crossover trial found NR reliably and significantly raised blood NAD+ in healthy middle-aged and older adults.

A separate crossover trial in healthy middle-aged and older adults confirmed NR was well tolerated and effectively raised blood NAD+ levels.
