Ergothioneine
The "longevity vitamin" with real, if early, human cognitive data
Ergothioneine is a naturally occurring antioxidant amino acid found in high concentrations in mushrooms — especially oyster and shiitake varieties. Humans and other animals can't synthesize it themselves; it has to come from diet. What makes it biologically unusual is that the body has a dedicated transporter, OCTN1, that actively pulls ergothioneine out of the blood and concentrates it in tissues under high oxidative stress, including the brain, eyes, and bone marrow.
That level of biological investment in a compound the body can't make is what earned ergothioneine its "longevity vitamin" nickname among researchers, most notably Barry Halliwell's lab in Singapore, which has spent over two decades studying its role in aging and cognitive health. Two small but real human randomized controlled trials have now tested it directly for cognitive outcomes, with genuinely encouraging results — though both are early-stage and modestly sized.
Cognitive Function & Memory
A 12-month pilot trial in adults with mild cognitive impairment found improved performance on a learning-and-memory test with ergothioneine versus placebo, alongside stabilized neurofilament light chain — a blood marker of neuronal damage that rose in the placebo group.
Antioxidant Protection
Unlike vitamin C or E, ergothioneine resists rapid metabolism and accumulates in high-stress tissues rather than being quickly cleared, giving it a longer window to neutralize free radicals where oxidative damage tends to concentrate.
Sleep Quality
The 16-week trial in older adults with subjective memory complaints also tracked sleep quality as a secondary outcome alongside its cognitive measures, part of a broader pattern of interest in ergothioneine's role in stress-tissue recovery.
Cellular Stress Resistance
Epidemiological research has linked lower blood ergothioneine levels to increased risk of cognitive decline and frailty in older adults, though association isn't the same as proof of a causal, correctable deficiency.
- Higher doses haven't shown clearly greater benefit in the trials so far — more isn't necessarily better here

A 2024 randomized, double-blind, placebo-controlled pilot study gave 19 adults with mild cognitive impairment either 25mg of ergothioneine three times weekly or placebo for one year. The ergothioneine group showed improved performance on a verbal learning test and stabilized neurofilament light chain levels, a marker of neuronal damage that significantly increased in the placebo group. A separate 2025 trial in 147 older adults with subjective memory complaints tested 10mg or 25mg daily for 16 weeks against placebo, with results published in the peer-reviewed journal Nutraceuticals.
